Most good stories end with the final, big conflict when the characters must choose what side of the book's main argument they're on. Then afterward, the main message of the story is really driven home.
Although for comedy, I suppose this would work a bit different. I like Talia Brie's comment about the punchline, and I think I'd have to agree there.
I'm not that great at comedy myself, so I'm probably not the best one to comment on this. But I wish you luck!
